Abstract : The quantification of epithelial barrier functions provides vital knowledge inunderstanding the physiological role of epithelial tight junctions in a multicellularorganism. Existing methods by measuring the trans-epithelial electrical resistance (TER)and using the Electrical Cell-substrate Impedance Sensing (ECIS) have proven to be verypromising but need to be improved for such problems as methodological inconsistencyand complexity. Our studies in this thesis work, by examining the electricalcharacteristics of epithelial cells on porous supports, are aimed to tackle these problems.The factor most complicating the extraction of epithelial barrier functions in themeasurements of TER and ECIS is the current flowing inside the narrow spaceunderneath cells. To explore the experimental approaches that can reduce or eliminatesuch a complication, we have constructed a mathematical model to analyze theimpedance of a celVelectrode system. This model singles out three cell-relatedparameters, a, RI, and C,: a stands for the impedance contribution from the above-mentioned current underneath cells, Rt, is an equivalent representation of epithelial barriertested for theQ1 PC1 bus cardBoth these projects mere sofixare des elopment efforts tonards contributing to dlfferentaspects of Roboucs and lZ1echatronics projects m the Controls and Roboucs Group..
