Abstract : This dissertation focuses on the design, development and applications of novelcomputational approaches for computer-assisted drug design (CADD) and protein modeling.The CADD approaches have been developed in the areas of both ligand-based and structure-based drug design. In addition, Simplicia1 Neighborhood Analysis of Protein Packing(SNAPP) has been employed for the analysis of protein flexibility.In the area of ligand-based design, I have implemented a new statistical modelingmethodology based on lazy learning theory (termed ALL-QSAR). To our knowledge, it isthe first application of this type of algorithm to QSAR studies. Compared with othermodeling approaches, ALL-QSAR can generate models with significantly higher predictivepower. This method has been successfully applied to hit identification for anticonvulsantagents via large chemical databasetested for theQ1 PC1 bus cardBoth these projects mere sofixare des elopment efforts tonards contributing to dlfferentaspects of Roboucs and lZ1echatronics projects m the Controls and Roboucs Group..
